بررسی مولکولی توان ضدمیکروبی ترشحات لارو مگس لوسیلیا سریکاتا علیه باکتری باسیلوس

با توجه به پیشرفت فن آوری برای بهبود زخم ها و مراقبت های پزشکی از آن ها هنوز هم زخم های nonhealthingبه علت گسترش باکتری¬های مقاوم به آنتی بیوتیک به صورت یک مشکل اساسی باقی مانده¬اند و سالانه هزینه های زیادی در این راه صرف می شود ،وباعث تمرکز در جستجوی درمان جایگزین برای آنتی بیوتیک شده است. واین به عنوان یک نیاز برای جامعه پزشکی محسوب می شود ، از لارو Lucilia sericataدر طی قرن¬ها برای بهبود زخم¬های مزمن استفاده ¬شده است. لارودرمانی یک برنامه کنترل شده است ،که از لارو Lucilia sericata استریل جهت بهبود زخم¬های مزمن استفاده می شود .در مطالعه میکروبی این رساله رشدباکتری B.subtilisمقاوم به آنتی بیوتیک در سه گروه عصاره لارو تیمارشده با باکتری ،عصاره لارو استریل و گروه کنترل بررسی شد،و رشدباکتری در بازه ی زمانی مختلف آنالیز شد و نتیجه گرفته شد که عصاره لارو تیمار شده با باکتری قادر است تا 2ساعت رشد باکتری B.subtilis مقاوم به آنتی بیوتیک را متوقف کند.وعصاره لارو استریل روی رشد باکتری تاثیر معنی داری ندارد .ودر بخش مطالعات مولکولی به بررسی تاثیر ترشحات لارو تیمارشده با باکتری B.subtilis بر روی ژن های PbpB وDnaB به عنوان ژن هدف در مقایسه با ژن gyrB که ژن مرجع است، پرداخته شد.و در نهایت برای بررسی بیان ژن¬های ذکر شده از روش Real time PCR استفاده شد . نتایج از این قرار بود،که بیان دو ژن هدف در مقایسه با ژن housekeeping به طور قابل توجهی کاهش داشته و این سبب مهار رشد باکتری B.subtilisمقاوم به آنتی بیوتیک شده است .پیشنهاد می گردد،که امکان دارد، ترشحات لارو پتانسیل تولید دارو را داشته باشند و امید است مشکلات مربوط به زخم مزمن برطرف گردد.

survey of antibacterial potential of Lucilia sericata larval secretion on Bacillus subtilis

With the advancement of technology for wound healing and medical care, nonhealthy wounds still remain a major problem due to the spread of antibiotic-resistant bacteria. And there's been a lot of spending this year, focusing on alternative antibiotic treatment options. This is considered a requirement for the medical community, The use of maggot therapy as a successful traditional therapeutic approach has again been considered.maggottherapy is a controlled program, used by Lucilia sericata larvae for the treatment of chronic ulcers. In this microbial study, the antibacterial resistant B.subtilis strain was studied in three groups of larvae treated with bacteria, larval sterile extract and control group, Growth bacteria were analyzed over time, It was concluded that larvae treated with bacteria can stop 2 hours of antibiotic resistant B.subtilis bacteria growth. And Sterile larvae have no significant effect on bacterial growth. In the molecular studies section, the effects of B.subtilis-treated larvae on the PbpB and DnaB genes as target genes were compared with the gyrB gene found in the housekeeping gene. Finally, real time PCR was used to determine the expression of these genes. The results were that the expression of two target genes was significantly reduced compared to the housekeeping gene, which in turn inhibited the growth of antibiotic resistant B.subtilis bacteria. It is suggested that larval secretions may have potential for drug production and hope to resolve chronic wound problems.
