شناسایی انواع سروتیپ های سالمونلا های جدا شده از مرغ های کشتار شده در شهرستان کاشمر به روش PCR و تعیین حساسیت آنتی بیوتیکی آنها

سالمونلوز یک بیماری مشترک بین انسان و حیوانات می باشد که توسط باکتری های جنس سالمونلا ایجاد می شود. سالمونلا باکتری گرم منفی، میله ای شکل و از خانواده انتروباکتریاسه می باشد. یکی از راه های مهم انتقال عفونت های سالمونلایی به انسان مصرف گوشت مرغ آلوده می باشد که باعث بروز بیماری های تب روده ای، باکتریمی و گاستروانتریت در مصرف کنندگان می شود. با توجه به جایگاه و اهمیت فراورده های طیور در سبد غذایی اقشار مختلف جامعه ، سلامت این محصولات دارای اهمیت زیادی می باشد. بنابراین این مطالعه، با هدف تعیین سروتیپ های سالمونلا و بررسی مقاومت آنتی بیوتیکی آنها در مرغ های کشتار شده در شهرستان کاشمر، انجام گرفت. بدین منظور تعداد 60 نمونه گوشت مرغ از فروشگاه های شهرستان کاشمر جمع آوری و جهت تعیین سروتیپ به آزمایشگاه منتقل شد. نمونه ها در محیط های غنی کننده ، مک کانکی و سالمونلا- شیگلا کشت داده شدند. نمونه های مشکوک جهت انجام تست های بیوشیمیایی در محیط های TSi و قند مانیتول کشت داده شدند و بر روی آن ها تست کاتالاز انجام شد. جهت شناسایی و تعیین سروتیپ نمونه های مشکوک، با استفاده از سه جفت پرایمر مربوط به ژن های invA، pefA و sefA، عمل PCR انجام شد. و پس از انجام PCR ، 7 نمونه سالمونلا شناسایی شد که 5 نمونه سروتیپ انتریتیدیس و یک نمونه سالمونلا تیفی موریوم بودند. در تعیین مقاومت آنتی بیوتیکی جدایه ها، بیشترین مقاومت نسبت به کانامایسین، لینکواسپکتین و کلرتتراسایکلین، و کمترین مقاومت نسبت به سیپروفلوکساسین دیده شد. با توجه به نتایج حاصله، مواد غذایی وابسته به طیور از مهمترین منابع باکتری سالمونلا می باشند و مصرف بی رویه آنتی بیوتیک در مرغداری ها باعث افزایش مقاومت در این پاتوژن شده است.

Identification of Salmonella serotypes isolated from poultry slaughtered in the city of Kashmar by PCR and determination of antimicrobial susceptibility.

Salmonellosis is a zoonotic disease in humans and animals caused by the bacteria Salmonella spp. Salmonella bacteria are gram-negative, rod-shaped and are of the family Enterobacteriaceae. One of the important ways of transmission of Salmonella infections in humans infected chicken meat consumption, which causes enteric fever, bacteremia and gastroenteritis in consumers. The consequences of poultry products in the food basket all walks of life, the health of these products is of great importance. This study aimed to determine the Salmonella serotypes and antibiotic resistance in poultry slaughtered in the city of Kashmar, was carried out. A total of 60 samples of chicken meat shop in the city of Kashmar collected and transferred to the laboratory to determine the serotype. Rich sample environments, the Enrichment, Macconkey and Salmonella-shigella were cultured. Suspicious samples for biochemical tests were cultured in an environment of TSi and sugar mannitol and catalase test was conducted on them. To determine the serotype suspicious samples, using three primer pairs of genes invA, pefA and seA, PCR was performed action. After the PCR, 7 of Salmonella enteritidis serotype 5 isolates were identified and a sample of Salmonella typhimurium. In determining antibiotic-resistant strains, most resistance to kanamycin, Lincospectin and Chlortetracyclin and was of least resistance to ciprofloxacin. According to the results depend on poultry for food, especially Salmonella enteritidis and Salmonella bacteria are the most important resources. And indiscriminate use of antibiotics in poultry has increased pathogen resistance
