ارزیابی مواد موثره عصاره گیاه انجبار بر زمان انعقاد خون

اختلالات انعقاد از جمله بیماری هایی است که افراد زیادی را درگیر کرده و مهمترین آنها هموفیلی است. هموفیلی به دلیل کمبود فاکتورهای انعقادی خون بوجود می آید که علت جهش در ژن های فاکتورهای انعقادی می باشد. هموفیلی در مردها شایع تر از زنان است و این افراد از عدم انعقاد خون رنج می برند. گیاهان دارویی در درمان بیماری های مربوط به انعقاد خون از قبیل هموفیلی همواره مورد توجه بوده اند. گیاه انجبار (Polygonum bistorta) در طب سنتی برای درمان بیماری های بسیاری به کار می رود. عصاره ریشه انجبار دارای اثرات ضد سرطانی، ضد التهابی، ضد میکروبی می باشد. این تحقیق با هدف بررسی اثر عصاره گیاه انجبار بر انعقاد خون انجام شد. برای این هدف ابتدا گیاه انجبار تهیه گردید. از ریشه گیاه انجبار به روش رفلاکس عصاره گیری شد و سه غلظت مختلف از عصاره تهیه شد. اثر غلظت های عصاره تهیه شده بر انعقاد خون مورد آزمایش قرار گرفت. برای این هدف، از سه تست زمان پروترومبین (PT) و زمان نسبی ترومبوپلاستین (PTT) و زمان لخته (CT)که فرایند انعقاد خون را بررسی می کند، استفاده گردید. برای انجام تست زمان پروترومبین و زمان نسبی ترومبوپلاستین از پول پلاسما استفاده گردید، در حالی که تست زمان لخته با خون فرد سالم که سابقه بیماری مرتبط با انعقاد خون نداشت، انجام شد. نتایج نشان داد که هر سه غلظت تهیه شده از عصاره آبی گیاه انجبار بر زمان پروترومبین اثر گذاشته و این زمان را در مقایسه با نمونه شاهد کاهش داند. نتایج مربوط به شاخص انعقادی زمان نسبی ترومبوپلاستین نیز نشان داد که عصاره آبی گیاه انجبار باعث کاهش زمان نسبی ترومبوپلاستین نسبت به نمونه شاهد گردید که بیشترین اثر این عصاره در غلظت 10/1 و سپس 20/1 و در نهایت 40/1 بود. نتایج مربوط به تست زمان لخته شدن (CT) نیز نشان داد که عصاره آبی گیاه انجبار منجر به کاهش زمان لخته شدن خون نسبت به نمونه شاهد شد. با توجه به نتایج بدست آمده از اثر گیاه انجبار بر سه تست انعقادی خون، میتوان عصاره آبی گیاه انجبار را به عنوان گزینه پیشنهادی در درمان بیماران هموفیلی در نظر گرفت

Evaluation of effective materials of blubber extract on coagulation time

Coagulation disorders are diseases that affect many people, most notably hemophilia. Hemophilia is caused by a lack of blood coagulation factors, which is the cause of mutations in the coagulation factor genes. Hemophilia is more common in men than in women, and these patients suffer from blood clotting. Medicinal plants have always been considered in the treatment of blood coagulation diseases such as hemophilia. Polygonum bistorta is used in traditional medicine to treat many diseases. Polygonum bistorta root extract has anti-cancer, anti-inflammatory, antimicrobial effects. The aim of this study was to investigate the effect of Polygonum bistorta extract on blood coagulation. For this purpose, an Polygonum bistorta plant was first prepared. The roots of the Polygonum bistorta were extracted by reflux method and three different concentrations of extract were prepared. The effect of extract concentrations on blood coagulation was investigated. For this purpose, three tests of prothrombin time (PT) and relative time of thromboplastin time (PTT) and clot time (CT) were used to evaluate the blood coagulation process. The plasma pool was used to perform prothrombin time and relative thromboplastin time, while the clot time test was performed with the blood of a healthy person with no history of coagulation-related disease. The results showed that all three concentrations of aqueous extract of Polygonum bistorta had effect on prothrombin time and decreased this time compared to control. The results of coagulation index of relative thromboplastin time also showed that the aqueous extract of Polygonum bistorta plant reduced the relative time of thromboplastin compared to control. The results of the clotting time (CT) test also showed that the aqueous extract of Polygonum bistorta plant reduced blood clotting time compared to the control sample. Based on the results of the effect of Polygonum bistorta on three blood coagulation tests, aqueous extract of Polygonum bistorta can be considered as a treatment option in hemophilia patients.
