بررسی خواص آنتی اکسیدانی،ضد میکروبی و شناسایی ترکیبات شیمیایی عصاره های حاصل از بخش های مختلف گیاه علف کبکی(Bongardia chrysogonum) و ارزیابی امکان استفاده از آن جهت افزایش پایداری اکسیداتیو روغن آفتابگردان.

اسانس ها و عصاره های حاصل از گیاهان دارویی با داشتن ترکیبات ضدمیکروبی، ضدسرطانی و آنتی اکسیدانی به عنوان ترکیبات جدید و طبیعی از اهمیت خاصی برخوردار می باشند. در مطالعه حاضر ترکیبات شیمیایی و خواص ضدمیکروبی عصاره های های حاصل از بذر، برگ و گل گیاه علف کبکی مورد بررسی قرار گرفت. سپس میزان کل ترکیبات فنلی و فعالیت مهار کنندگی رادیکال های آزاد عصاره های استخراج شده در غلظت های مختلف ( 250، 500 و 1000پی پی ام) بررسی گردید و با قدرت رادیکال گیرندگی آنتی اکسیدان سنتزی BHA در سطح 100 پی پی ام مقایسه شد. در مرحله بعد غلظت های مختلف عصاره بهینه حاصل از بخش های مختلف گیاه علف کبکی به روغن آفتابگردان تصفیه شده فاقد آنتی اکسیدان افزوده شد و پایداری اکسایشی روغن آفتابگردان به مدت سه روز از طریق سنجش اندیس پراکسید، عدد اسیدی، شاخص تیوباربیتوریک اسید و طول دوره القاء بررسی و در نهایت با فعالیت آنتی اکسیدان سنتزی BHA به میزان 100 پی پی ام مقایسه گردید. نتایج نشان دادند که میزان آلفاتوکوفرول در عصاره برگ گیاه علف کبکی به میزان قابل ملاحظه ای بیشتر از دو بخش بذر و گل بود و میزان بتا و گاماتوکوفرول در عصاره گل بیشتر مشاهده شد. مقادیر کل استرول ها و ترکیبات فنلی در بخش عصاره بذر بیشتر از بخش های برگ و گل گیاه بود. نتایج فیتوشیمی نشان داد که ترکیباتی از قبیل استروئیدها، ترپن ها، تانن ها و آلکالوئیدها در بخش های مختلف گیاه علف کبکی وجود دارند. آنالیز میکروبی نشان دادند که عصاره بذر بر باکتری های گرم منفی، عصاره گل بر باکتری های گرم مثبت و عصاره برگ بر قارچ ها موثرتر بودند و به طور قابل ملاحظه ای توانسته اند رشد میکروارگانیسم ها را محدود نمایند. با افزایش غلظت عصاره بذر، برگ و گل گیاه علف کبکی میزان ترکیبات فنلی، آنتی اکسیدانی و فعالیت مهارکنندگی رادیکال های آزاد افزایش یافت و غلظت 1000 پی پی ام عصاره بذر گیاه علف کبکی دارای بالاترین قدرت آنتی اکسیدانی نسبت به سایر غلظت ها بود. بنابراین عصاره بذر گیاه علف کبکی به عنوان عصاره بهینه آنتی اکسیدانی حاصل از بخش های مختلف گیاه علف کبکی (برگ، گل و بذر) انتخاب گردید و به روغن آفتابگردان تصفیه شده فاقد آنتی اکسیدان اضافه گردید. نتایج حاصل از بررسی پایداری اکسیداتیو روغن آفتابگردان نشان داد که غلظت 1000 پی پی ام عصاره بذر گیاه علف کبکی در کاهش اندیس پراکسید، شاخص تیوباربیتوریک اسید و عدد اسیدی و هم چنین افزایش شاخص پایداری اکسایشی روغن آفتابگردان نسبت به سایر غلظت های عصاره، نمونه شاهد و آنتی اکسیدان سنتزی BHA در سطح 100 پی پی ام تأثیر بیشتری داشته است. طبیعتاً قابلیت آنتی اکسیدانی بالاتر غلظت ppm1000 عصاره بذر گیاه علف کبکی طی فرآیند حرارتی را می توان به مقادیر باقی مانده ترکیبات فنلی و توکوفرولی موجود در آن در دمای 60 درجه سانتی گراد و هم چنین مقادیر بالای این ترکیبات آنتی اکسیدانی در عصاره نسبت داد. بدین ترتیب می توان عصاره بذر گیاه علف کبکی را در سطح 1000 پی پی ام به عنوان منبع مناسبی برای آنتی اکسیدان های طبیعی و ترکیبات ضد میکروبی معرفی نمود.

Study of Antioxidant and Antimicrobial Properties and Recognition of Chemical Compounds of Extracts from Various Parts of Bongardia chrysogonum and Propability Using that for Improvement of Oxidative Stability of Sunflower Oil.

Essential oils and extracts obtained from medicinal plants enjoy special importance as new and natural products because of their antimicrobial, anticancer, and antioxidant compounds. The present research studied the chemical compounds in and antimicrobial effects of seed, leaf, and flower extracts of Bongardia chrysogonum.total phenol contents of the extracts and their inhibitory activities at various concentrations (250 ppm, 500 ppm, and 1000 ppm) on free radicals were then determined and compared with the radical scavenging power of the synthetic antioxidant BHA at 100 ppm. In the next stage, different concentrations of the optimum extract obtained from the various parts of the plant were added to purified sunflower oil that did not contain antioxidants, and oxidative stability of the oil was investigated for three days through assessing the peroxide, acidic, and thiobarbituric acid values and the length of the induction period and finally, compared with the antioxidant activities of the synthetic antioxidant BHA at 100 ppm. Results indicated that the alpha-tocopherol content of the leaf extract was considerably greater than those of the seed and flower extracts, whereas the beta and gamma-tocopherol contents of the flower extract were higher. The total contents of sterols and phenolic compounds in the seed extract were higher than those of the leaf and flower extracts. Results of phytochemical analysis showed that there were compounds including steroids, terpenes, tannins, and alkaloids in the various parts of this plant. Microbial analysis revealed that the seed, the flower, and the leaf extracts were more effective on Gram-negative bacteria, Gram-positive bacteria, and fungi, respectively, and were able to considerably limit growth of these microorganisms. Increases in the concentrations of the seed, leaf, and flower extracts of the plant improved their contents of phenolic compounds and their antioxidant and inhibitory activities on free radicals, and the 1000 ppm concentration of the seed extract of the plant had the highest antioxidant power among the studied concentrations. Therefore, the seed extract was selected as the optimum antioxidant extract obtained from the various parts (leaf, flower, and seed) of the plant and added to the purified sunflower oil that did not contain antioxidants. Results of studying oxidative stability of sunflower oil demonstrated that the seed extract of the plant at 1000 ppm was more effective than its other concentrations, the control sample, and the synthetic antioxidant BHA in reducing peroxide, thiobarbituric acid, and acid values and also in increasing the oxidative stability index of the sunflower oil. Naturally, the higher antioxidant capacity of the seed extract at 1000 ppm during the thermal process can be attributed to the remaining amounts of phenolic compounds and of tocopherols present in it at 60?C and also to the large quantities of these antioxidant compounds in the extract. Therefore, we can introduce the seed extract of Bongardia chrysogonum at 1000 ppm as a suitable source of natural antioxidants and of antimicrobial compounds
