پیش بینی خاصیت ضدمیکروبی پوشش خوراکی تولید شده از نشاسته حاوی عصاره الکلی پوست انار با روش الگوریتم ژنتیک

در این تحقیق اثر ضد میکروبی فیلم نشاسته حاوی غلظت های مختلف عصاره الکلی پوست انار بر باکتری های لیستریا مونوسیتوژنز، اشرشیا کلی، استافیلوکوکوس اورئوس، سالمونلا تیفی موریوم، سودوموناس ایروژینوزا و باسیلوس سرئوس مورد بررسی قرار گرفته، ویژگی های مکانیکی و فیزیکی فیلم های تهیه شده مطالعه گردید و به پیش بینی خاصیت ضدمیکروبی پوشش های خوراکی تهیه شده با روش الگوریتم ژنتیک پرداخته شد. برای بررسی خواص ضد میکروبی از روش انتشار از دیسک استفاده گردید. از فیلم های نشاسته دیسک هایی به قطر 10 میلی متر تهیه گردید و بر روی محیط های کشت BHI آگار که قبلا با هر یک از باکتری ها تلقیح شده بودند قرار گرفت. قطر هاله های مهاری به عنوان شاخص میزان فعالیت ضد میکروبی فیلم ها در نظر گرفته شد. میزان نفوذپذیری فیلم ها در برابر بخار آب با دستورالعمل اصلاح شده ای از روش E96-95 مصوب ASTM محاسبه گردید. خواص کششی فیلم ها نیز با دستگاه سنجش کشش و روش D882-91 مصوب ASTM بررسی شد. محاسبات آماری با نرم افزار SPSS و در قالب طرح کاملا تصادفی انجام و مقایسه میانگین ها از آزمون چند دامنه ای دانکن صورت گرفت. جهت تخمین بهینه ترین مقادیر برای پارامترهای دما، pH، قطر هاله ضدمیکروبی و غلظت، کروموزوم مربوطه تشکیل و با توجه به داده های آزمایشگاهی میزان برازش هر یک از کروموزوم ها محاسبه و امتیاز آن کروموزوم تعیین گردید. نتایج نشان داد که فیلم های حاوی عصاره در غلظت 5% و بالاتر بر تمام باکتری های مورد مطالعه دارای خاصیت مهاری بود هر چند که غلظت 1% عصاره باعث مهار سودوموناس ایروژینوزا، باسیلوس سرئوس و استافیلوکوکوس اورئوس گردید. افزودن عصاره به فیلم نشاسته و افزایش غلظت آن سبب بالا رفتن معنی دار میزان نفوذپذیری فیلم ها نسبت به بخار آب، کاهش مقاومت کششی و افزایش ازدیاد طول گردید. با توجه به ضرایب همبستگی محاسبه شده بین مقادیر واقعی و مقادیر پیش گویی شده، مدلسازی به روش الگوریتم ژنتیک بعنوان یک روش مناسب جهت پیش بینی خاصیت ضدمیکروبی پوشش های خوراکی تولید شده از نشاسته حاوی عصاره الکلی پوست انار بر روی شش نوع باکتری مورد مطالعه ارزیابی شد.

Prediction of antimicrobial properties of starch based edible coating incorporated with alcoholic extract of pomegranate rind with genetic algorithm

In this study, the antimicrobial activity of starch based edible film incorporated with different concentration of pomegranate rind alcoholic extract against Listeria monocytogenes, Escherichia coli, Staphylococcus aureus, Salmonella typhimurium, Pseudomonas aeroginusa and Bacillus cereus were surveyed, mechanical and physical properties of the films were studied, and antimicrobial activity of developed edible coatings was predicted by genetic algorithm. The disc diffusion method was used for determination of the antimicrobial effect of films. Starch films were cut to the discs with 10 mm diameter and were placed in to BHI Agar medium which had been previously seeded with tested bacteria. The inhibitory activity was measured based on the average diameter of the inhibitory zone. The water vapor permeability of films was determined using a modified ASTM E96-99 procedure. Tensile properties of the films were determined using a tensile tester according to ASTM standard method D882-91. Statistics on a completely randomized design were performed in SPSS software and Duncan’s multiple range tests was used to detect differences among mean values. To determine the optimal values for the temperature, pH, concentration and inhibition zone, corresponding chromosomes formed and according to experimental data the fitness of each chromosome calculated and chromosome were rated. The results show that all films containing 5% and higher concentration of extract inhibited the growth of tested bacteria, although the films containing 1% of extract inhibited the growth of P. aeroginusa, B. cereus and S. aureus. The addition of extract to starch films and increasing of its concentration significantly increased WVP, decreased tensile strength and increased elongation. According to the calculated correlation coefficient between actual and predicted values, genetic algorithm modeling were determined as a suitable method for forecast antimicrobial activity of edible coatings produced from starch-containing alcoholic extract of pomegranate peel on six types of bacteria.
