بررسی بیان ژن های oct۴ و sox۲ در ناخنک چشم

به حروف : خلاصه پایان نامه (چکیده پایان نامه در حد اکثر 250 کلمه به زبان فارسی ) هدف : بیماری ناخنک چشم رشد خوش خیم بافت فیبروواسکولار روی قرنیه است. ناخنک چشم یک بیماری سطح چشم است که توسط تکثیر غیرطبیعی اپیتلیال، تغییر شکل ماتریکس، رگ زایی و مهاجرت آسیب شناخته می شود. اگرچه اتیولوژی بیماری ناخنک چشم وسیع است، اما مطالعات اخیر بر روی نقش آسیب سلول های بنیادی لیمبال (LSCs) و تأثیر UVB متمرکز شده اند. هدف از این مطالعه بررسی سطح بیان ماکرهای پرتوان در بیماران دارای ناخنک چشم است. مواد و روش ها : با استفاده از روش Real Time PCR بیان SOX2 و OCT4 در ناخنک چشم اولیه در مقایسه با بافت طبیعی ملتحمه مورد بررسی قرار گرفت. ارتباط بین بیان SOX2 و OCT4 و همچنین ارتباط بین فاکتورهای کلینیکوپاتولوژیکال با هر دو بیان ژن ارزیابی شد. نتایج : سطح بیان نسبی ژن های OCT4 در بافت ناخنک اولیه، کاهش معنی داری را در مقایسه با بافت ملتحمه نرمال نشان داد. ارتباط بین بیان ژن OCT4 و فاکتورهای کلینیکوپاتولوژیکال در یکطرفه/دوطرفه بودن معنی دار (p=0.04)، و نیز به صورت مرزی با فاکتور فعالیت رشد معنی دار است (p=0.063). در نهایت بین بیان ژن SOX2 و OCT4 همبستگی یک طرفه وجود دارد (p=0.001). بحث و نتیجه گیری: نتایج این مطالعه، کاهش بیان نشانگرهای پرتوانی، ژن OCT4 را در ناخنک چشم نشان می دهد. به نظر می رسد که این نتایج ممکن است یک راه جدیدی را در مشمول بودن LSCs در ناخنک چشم پیش بینی کند.

Expression analysis of the SOX2, OCT4genes in pterygium

Pterygium is a benign growth of fibrovascular tissue on the cornea. It is a common ocular surface disease characterized by the abnormal epithelial proliferation, matrix remodeling, vascularization and the migration of the lesion. Although the etiology of pterygium is elusive, recent studies have focused on the role of limbal stem cells (LSCs) damage and effects of UVB. The aim of this study was to examine the expression levels of pluripotent markers in petrygium patients. Methods: Using real time PCR, the expression of SOX2 and OCT4 were analyzed in primary pterygium when compared to their normal conjunctiva tissues. The correlation between SOX2mRNA expressions with OCT4 mRNA expression, as well as association between clinicopathological indices with both gene expression levels was assessed. Results: The relative mRNA expression levels of OCT4 genes in primary pterygium tissues revealed significant decreased compared with normal conjunctiva tissues. The association between OCT4 gene expression and clinicopathological indices were significant in laterality (p= 0.04), and were marginally significant with growth activity (p= 0.063) indices. Finally, the univariate correlation between expression of SOX2 and expression of OCT4 were significant (p= 0.001). Conclusion: The outcomes of present data highlighted the down-regulation of pluripotency markers OCT4 genes, in the pterygium. It is speculated that these results may be predicted a new avenue for the involvement of LSCs in pterygium
