استخراج عصاره متانولی گل خارمریم (Silybum Maryiamnum) و بررسی اثرات آنتی اکسیدانی و پایدار کنندگی آن در روغن سویا در طی نگه داری

برای جلوگیری از اکسیداسیون روغن ها روش های متعددی وجود دارد که یکی از این موارد افزودن آنتی اکسیدان های سنتزی می باشد.اما با توجه به این که آنتی اکسیدان های سنتزی اثرات نامطلوبی هم چون اثر جهش زایی و سرطان زایی در بدن انسان دارند،لذا تحقیق و بررسی منابع آنتی اکسیدان طبیعی به منظور جایگزین کردن ترکیبات سنتزی ضروری به نظر می رسد.در این پژوهش ابتدا عصاره متانولی گل گیاه خارمریم استخراج گردید.سپس عصاره استخراج شده در غلظت های مختلف ( 500 – 600 - 700 – 800 – 900 پی پی ام) به روغن سویا تصفیه شده فاقد آنتی اکسیدان افزوده شد و همچنین پایداری اکسایشی نمونه ها در دمای 65 درجه سانتیگراد به مدت 3 روز از طریق اندازه گیری اندیس پراکسید،اندیس تیوباربیوتیک اسید و اندیس اسیدی و در نهایت با فعالیت آنتی اکسیدان سنتزی BHT به میزان PPM 200 مقایسه گردید.به طور کلی نتایج نشان داد با افزایش غلظت عصاره گل گیاه خارمریم در روغن سویا از 500 تا 900 پی پی ام در یک زمان نگه داری ثابت،شاخص پایداری اکسایشی ،میزان ترکیبات فنلی و فعالیت مهارکنندگی رادیکال های آزاد در روغن افزایش یافته در حالی که اندیس TBA کاهش پیدا کرد.نتایج حاصل از بررسی پایداری اکسایشی روغن حاوی غلظت های مختلف عصاره نشان داد غلظت900 پی پی ام عصاره نسبت به غلظت های دیگر و نمونه شاهد به دلیل داشتن مقادیر بالاتر ترکیبات پلی فنلی و توکوفرولی در پایداری اکسایشی روغن سویا موثرتر عمل نموده و در مقایسه با آنتی اکسیدان سنتزی BHT در غلظت 200 پی پی ام نیز تاثیر بیشتری داشت. بدین ترتیب می توان گل گیاه خارمریم را در سطح 900 پی پی ام به عنوان منبع مناسبی برای آنتی اکسیدان های طبیعی معرفی نمود.

Extraction Silybum Maryiamnum Flower and Study of The Antioxidant Effect and Stability on Soybean oil in Storage

There are many ways of preventing oxidation of oils, one of which is the addition of synthetic antioxidants to them. However, given the fact that synthetic antioxidants have adverse effects , such as mutagenic and carcinogenic effects , on the human body, it seems necessary that research and studies should be carried out with the purpose of replacing synthetic antioxidants. In this research, we first obtained methanol extracts of Silybum Maryiamnum flower and identified their constituents. The extracts were then added at various concentrations (500-600-700-800 and 900ppm) to purified antioxidant-free soyben oil. The inhibition of the activity of the free radicals by the extracts and the oxidative stability of the oil at 65 degrees centigrade were studied for seven days by measuring the peroxide index, the TBA index, the number of conjugated diene, and the duration of induction. Finally, the activities of the extracts were compared with those of the synthetic antioxidant TBA that, at the concentration of 200ppm, had been added to the purified canola oil. In general, results obtained showed that addition of 500 to 900 ppm extracts of Silybum Maryiamnum flower to soyben oil at a given storage time increased the oxidative stability index, the quantities of polyphenolic compounds, and the inhibition of the activities of the free radicals in the oil, but decreased the peroxide index, the TBA index, and the number of conjugated diene. On the other hand, at a given concentration of the extracts, the peroxide index, the TBA index, and the number of the DNA conjugates increased by increasing the storage period of the oil from one to seven days, but the duration of induction decreased. Therefore, results obtained from investigating the oxidative stability of the oil containing various concentrations of the extracts indicated that extracts at the concentration of 200 ppm, as compared with the other concentrations of extracts and the control sample, were more effective with respect to the oxidative stability of soyben oil. The reason for this was that they contained more of polyphenolic and tocopherol compounds, and were more effective than the BHT synthetic antioxidant added at the concentration of 200 ppm to the oil. Therefore, extracts of Silybum Maryiamnum flower at the concentration of 900ppm can be introduced as a suitable replacement for synthetic antioxidants.
