بررسی تولید آنزیم آلفا آمیلاز توسط باکتری های هالوفیل جداشده

آلفا آمیلازها از مهمترین و با اهمیت ترین آنزیم ها به شمار می آیند و اهمیت فراوانی در بیوتکنولوژی امروزی ایفا می کنند. اگر چه منابع مختلفی قادر به تولید آلفا آمیلاز می باشند عموما آنزیم های مشتق شده از منابع میکروبی به ویژه منابع باکتریایی و از جنس باسیلوس از جنبه های کاربردی و صنعتی دارای ارزش می باشند و بیشتر مورد مطالعه و بررسی قرار گرفته اند. در این تحقیق ابتدا سویه های باکتری از مناطق نمک دوست جداسازی و نسبت به تولید آنزیم تجزیه کننده نشاسته غربالگری شدند. در این مطالعه سویه باکتریای مقاوم به نمک توانایی تولید آلفا آمیلاز خارج سلولی را دارا بود.در ادامه با استفاده ازآنالیز16S rDNA و با رسم درخت فیلوژنتیکی جایگاه این سویه در مقایسه با سایر باسیلوس های شناخته شده تعیین شد. مطالعات تعیین توالی نشانگر شباهت بالای سویه با سویه Bacillus sp. بود. با استفاده از کروماتوگرافی تعویض یونی و بکار بردن ستون Q-sepharose آلفا آمیلاز حاصل از Bacillus sp. خالص و سپس تعیین خصوصیت گردید. فعالیت آنزیمی به کمک روش برنفلد و در حضور بافر تریس Mm 50 تعیین شد. این آنزیم برای فعالیت به کلسیم نیاز ندارد. دما و pH بهینه این آنزیم به ترتیب 50درجه سانتی گراد و4/7 بود. الکتروفورز با SDS-PAGE باندی را با وزن مولکولی58 کیلو دالتون نشان داد. فعالیت این آنزیم در حضور درصد های مختلف نمک مثل کلرید سدیم و کلرید منیزیم بررسی شد. آنزیم در حضور60% از کلرید سدیم بطور میانگین تا 70% از فعالیت خود را کاملا حفظ کرده و در حضور40% از کلرید منیزیم 67 درصد از فعالیت در آنزیم باقی ماند. پایداری حرارتی آنزیم در حضور کلسیم , کلرید سدیم و کلرید منیزیم بررسی گردید. این آنزیم در حضور یونهای منگنز و آلومینیوم دچار کاهش فعالیت گردید، در حضور یون باریوم و لیتیوم تغییر شاخصی در فعالیت مشاهده نمیشود.

Production identification alpha amylaz enzyme by halophilic isolated

Alpha¬ amylase of the most important enzymes and play an important role in modern biotechnology. Although a variety of sources able to generate alpha¬amylase, Generally enzymes derived from microbial sources, particularly sources of bacteria of the genus Bacillus include valuable aspects. Furthermore industrial applications And more are being studied. Firstly, the bacterial strains isolated from industrial halophilic areas and starch degrading enzymes and were screened for the production of enzyme. In this study, strains of resistant halophilic bacteria capable of producing extracellular alpha -amylase respectively. The analysis of the 16S rDNA of these strains was recorded and the phylogenetic tree of the isolating were determined in comparison with other well-known bacillus. Sequencing studies showed 99% similarity with the strain Bacillu sp. Also ion exchange chromatography using Q-sepharose column alpha - amylase from Bacillus sp. net and then set the properties. Enzyme activity in the presence of Tris buffer 50 Mm by Bernfeld method was determined . The enzyme does not require calcium for activity. Temperature and pH optimum of the enzyme, were respectively, 50 ° C and 7.4 respectively. SDS-PAGE electrophoresis with molecular weight band 58 KDa showed. The enzyme activity in the presence of various salts such as Nacl and MgCl2 were reviewed. 70% of the enzyme activity is remined in the presence of NaCl (60%) and on average 67% of its activity is fully retained in the presence of 40% MgCl2. The thermal stability of isolated enzyme in the presence of calcium, NaCl and MgCl2 was investigated. The enzyme activity in the presence of ions, Mn+2 and Al+3 decreased but in the presence of ions Ba+2 and Li+1 no significant changes showed in activity.
